Transmission Electron Microscopy (TEM) and Scanning Electron Microscopy (SEM)

We have successfully conducted the first round of SEM and TEM of the L. monocytogenes before and after high pressure. We used three different fixative protocols for the SEM and for the TEM.

In the images, we can clearly see the membrane, and damaged Listeria cell after appyling high pressure.

All together, we have taken 8 experiments with over 100 images, with enlargement of up to x100.000 ! We are currently analyzing the results. These will be combined with FTIR and other methods to understand the damage to the listeria following high pressure stress.
